一般說明
细胞死亡（尤其是凋亡）是一个能量依赖的过程，需要ATP。随着ATP水平跌倒到细胞无法再执行基本代谢功能的程度，细胞将死亡。典型的凋亡细胞表现出ATP水平的显著降低。因此，细胞中ATP水平的损失已被用作细胞死亡的指标。相反，ATP水平升高已成为细胞增殖的指标。Calbiochem®品牌的ATP分析试剂盒利用ATP水平的生物发光检测，可同时快速筛选哺乳动物细胞中的凋亡和细胞增殖。该测定法利用荧光素酶催化ATP和荧光素形成光，并且可以使用发光计或β计数器测量光。
该测定基于在ATP和氧气存在下萤火虫荧光素酶催化的D-荧光素氧化，从而通过产生的光量（hv）定量ATP的量。

原則
该测定法可全自动测定高通量（10 s/样品），且灵敏度极高（可检测10-100个哺乳动物细胞/孔）。该测定法的高灵敏度已导致许多其他应用，用于检测各种酶促反应中的ATP产生，以及检测样品（例如血液、牛奶、尿液、土壤和污泥）中的低水平细菌污染。
準備報告
试剂复溶1.用2 ml酶复溶缓冲液复溶ATP监测酶。缓慢移液混匀。复溶酶在4°C下可稳定保存2个月。2.将1 mg ATP溶于1 ml H2O中，制备ATP标准溶液。该溶液可在-20°C下稳定保存几周。
