Desmin (UniProt: P17661) is encoded by the DES (also known as CDCD3, CMD1F, CSM1) gene (Gene ID: 1674) in human. Desmin is a homopolymeric protein that is classified as a muscle-specific type III intermediate filament. It is shown to be essential for proper muscular structure and function. It plays a crucial role in maintaining the structure of sarcomeres, inter-connecting the Z-disks and forming the myofibrils, linking them not only to the sarcolemmal cytoskeleton, but also to the nucleus and mitochondria, thus providing strength for the muscle fiber during activity. It also acts as a sarcomeric microtubule-anchoring protein that specifically associates with detyrosinated tubulin-alpha chains, leading to buckled microtubules and mechanical resistance to contraction. Desmin can undergo phosphorylation at multiple serines by CDK1. Phosphorylation at serine 60 by Aurora B and threonine 76 by ROCK1 is reported to contribute to efficient separation of Desmin intermediate filaments during mitosis. Mutations in DES gene have been linked to myofibrillar myopathy that is characterized at ultrastructural level by disintegration of the sarcomeric Z disk and myofibrils, and replacement of the normal myofibrillar markings by small dense granules, or larger hyaline masses, or amorphous material. It leads to skeletal muscle weakness associated with cardiac conduction blocks, arrhythmias, and restrictive heart failure. Application

Anti-Desmin, clone 1H16 ZooMAb , Cat. No. ZRB1178, is a highly specific rabbit recombinant monoclonal antibody that targets Desmin and has been tested for use in Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Desmin in lysates from C2C12 cells and rat heart tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Desmin in human skeletal muscle and rat heart tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Desmin in human skeletal muscle cells (SkMC) and C2C12 cells.

Note: Actual optimal working dilutions must be determined by the end user as specimens and experimental conditions may vary.

Biochem/physiol Actions

Clone 1H16 is a ZooMAb , rabbit recombinant monoclonal antibody that specifically detects Desmin. It targets an epitope within the C-terminal region.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose. Normal appearance is a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
