General description

Silymarin is a flavonolignan, obtained from milk thistle (Silybum marianum) plant.

Application

Silymarin has been used to study:
  • its in vitro antiviral, antibacterial, antifungal activities and cytotoxicity
  • its effect of silymarin on bladder contractions in cyclophosphamide (CYP)-induced cystitis rat model
  • its effect on liver toxication induced by Fumonisin B1 in mice

Biochem/physiol Actions

Silymarin is originally known to protect liver from various toxic agents and is used in treating hepatitis and cirrhosis. Silymarin possesses many biological actions such as antifibrotic, anti-lipid peroxidative, anti-inflammatory, immunomodulatory and membrane stabilizing. Silymarin also aids in liver regeneration.
Silymarin provides cardioprotective activity against ischemia-reperfusion induced myocardial infarction in rats.
Silymarin was shown to protect the liver from the cytotoxic effects of anti-tuberculosis drugs by decreasing serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP) levels. This effect was related to the anti-oxidant effects of silymarin.

Other Notes

Mixture of anti-hepatotoxic flavonolignans from the fruit of Silybum marianum.

Questions

1–5 of 5 Questions
  1. WHAT IS THE MOLECULAR WEIGHT? cAN i DISSOLVE IT IN dmso, i WANT TO USE IT IN CELL LINES?

    1 answer
    1. This product contains a mixture of anti-hepatotoxic flavonolignans from the fruit of Silybum marianum, and the molecular weights and distribution of this mixture have not been characterized. Although not tested for batch release, this compound is known to dissolve in DMSO at least up to a concentration of 20 mM, as referenced in Cellular Signalling 84 (2021): 110023.

  2. This silymarine.....is it isomer of Silybinin A and B

    1 answer
    1. This product is a fairly crude extract from Milk Thistle consisting of a number of components. The minimum purity specification for this material is 30%. Within that fraction there is a mixture of isomers. The amount of each has not been quanitified, however the literature suggests that these are present in the plant at equimolar ratios. Please see the link below to review a sample or lot specific Certificate of Analysis:

  3. How much purity is there in this product?

    1 answer
    1. This product is at least 30% Silymarin. The impurities have not been fully characterized. However, other Milk Thistle flavonolignans are expected to be present. These include dehydrosilybin, desoxysilycristin, desoxysilydianin, silandrin, silybinome, silyhermin, and neosilyhermin. In addition, apigenin, silybonol, as well as myristic, palmitic, stearic, and oleic acids have also been reported.

  4. I want to give it orally to a mouse. How should I prepare it?

    1 answer
    1. This product has not been tested for in vivo or in vitro use. The solubility of this compound is tested at 10 mg/mL in ethanol, which may not be ideal for animal models. There are numerous published papers noting oral administration in mice.

See the links below to review examples:
      https://www.ncbi.nlm.nih.gov/pmc/articles/PMC5156955/

  5. how can we dissolve silymarin ( in vitro study )

    1 answer
    1. The solubility of this material is tested at a concentration of 50 mg per 5 mL of ethanol, giving a clear to slightly hazy appearance.
