General description

Lactate is generated by lactate dehydrogenase (LDH) under hypoxic or anaerobic conditions. Monitoring lactate levels is, therefore, a good indicator of the balance between tissue oxygen demand and utilization and is useful when studying cellular and animal physiology. Simple, direct and automation-ready procedures for measuring lactate concentration are very desirable.

Biochem/physiol Actions

The Lactate Assay Kit is based on lactate dehydrogenase catalyzed oxidation of lactate, in which the formed NADH reduces a formazan (MTT) reagent. The intensity of the product color, measured at 565 nm, is proportionate to the lactate concentration in the sample.

  1. How the blood samples must be collected? Specific tube? How they must be conserved and for how long? Should we use serum or plasma? Thank you

    1. Both serum and plasma collected with EDTA or heparin are compatible with this assay. Whole blood should be processed into serum or plasma and frozen as quickly as possible if not tested immediately, as glycolysis will continue in blood after collection, increasing lactate concentration. Frozen samples should be kept at -20 °C or colder and can be stored for up to one month. Plasma may be favored since waiting for blood to clot is unnecessary, reducing the time for lactate accumulation.

  2. Can I measure lactate concentrations higher than 2 mM with this kit by just extending the calibration curve? I need to cover higher concentrations, around 4-5 mM. Thanks!

    1. It is recommended to run samples with different dilutions to ensure readings are within the standard curve rather than extending the standard curve to measure samples at higher concentrations. The results can then be multiplied by the dilution factor as advised in the technical bulletin.

  3. I need to do a L-Lactate time course in a 7-days fermentation, can I freeze my media samples?

    1. Although it is generally recommended to test fresh samples, it is likely that media samples can be frozen. But in order to confirm the validity of freezing samples, you might want to do the following:
      Assuming that samples will be taken once every day over those seven days, remove duplicate samples on days 1 and 4. Freeze one sample, and test the other. Then, on day 7, test the frozen samples from all seven days, and see how the frozen samples from days 1 and 4 compare to the fresh samples tested on those days. That way you will only need to run assays on three of the seven days.

  4. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  5. I have medium supernatant. The kit instructs not to add the enzyme to the blank, but it shows a different color in the culture medium. Should I include the enzyme in the blank? Also, should I use medium for calibration instead of the provided buffer?

    1. Two reactions are run when there is endogenous enzyme activity : one with added Enzyme A and a control reaction with no Enzyme A. The reaction mix preparation for the sample control does not include enzyme A.

      The initial dilution of the 20 mM standard to 2 mM is a 10x dilution that uses ultra-pure water as the solvent. The subsequent dilutions of the standard for a cell culture sample without phenol red is prepared in water. If the culture media contains phenol red the standard should be diluted directly in the culture media without serum as described by table 2 in the technical bulletin. Decide which preparation to follow based on whether the sample contains or does not contain phenol red.

  6. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.   7. For the lactate concentration standard curve, lactate standard solutions have to be prepared. Can these solutions be frozen and used multiple times? This would save a lot of time per Assay.

    1. The L-Lactate standard provided in this kit is prepared as a concentrated solution at 20 mM solution. Following dilution to the recommended working concentration the solution may not be stored. It is best prepared fresh.
