General description

Autophagy is an evolutionarily conserved degradation process that targets long-lived proteins, organelles, and other cytoplasmic components for degradation via the lysosomal pathway. Activation of the autophagy pathway is required for multiple cellular roles, including survival during starvation, the clearance of intracellular components, development, and immunity.

Application


  • Inhibitory Effect of Ursolic Acid on Proliferation and Migration of Renal Carcinoma Cells and Its Mechanism.: This study explores the inhibitory effects of ursolic acid on the proliferation and migration of renal carcinoma cells. The use of an autophagy assay kit was instrumental in demonstrating that ursolic acid induces autophagy, leading to decreased cell viability and migration. These findings highlight the potential of ursolic acid as a therapeutic agent in renal carcinoma through the modulation of autophagy (Lyu et al., 2022).

This kit is suitable for the detection of autophagy in mammalian cells by fluorescence microscopy and fluorescence microplate readers.

The Autophagy Assay kit provides a simple and direct procedure for measuring autophagy in a variety of cell types using a proprietary fluorescent autophagosome marker (λex = 333/λem = 518 nm).

  2. Can this autophagy assay kit detect lysosomes alone that are not merged with autophagosomes?

    1. This product is not designed to detect lysosomes alone that are not merged with autophagosomes. The fluorescent marker specifically labels autophagosomes and autophagic vacuoles, not lysosomes in isolation.

  3. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  4. if I have cells expressing GFP, Can I detect autophagy without the signal overlapping?

    1. This kit operates with an λex = 333/λem = 518 nm as the probe in this kit is a blue fluorescent dye. It is our understanding that GFP has an excitation of 395 nm with an emission at 508 nm. It is unclear if there is any signal overlap and this study would need to be determined empirically in a laboratory setting.

  5. The autophagy assay kit MAK138 says that it "measures autophagy by using a proprietary fluorescent autophagosome marker" However, it is not clear as to what is this marker? Where does this marker bind on the autophagosome? Does it bind to LC3II?

    1. The Autophagosome Detection Reagent used in the kit is a fluorescence marker that can label the acidic autophagosomes. It can be accumulated in autophagic vacuoles via both ion trapping and specific interactions with vacuole membrane lipids.

    1. The kit can detect whether an autophagosome has merged with a lysosome. The kit cannot distinguish between autophagy initiation and autophagy completion.

  7. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.
