General description

Chromium, an essential trace element, is a transition metal that primarily exists in two stable oxidation states, hexavalent [Cr(VI)] and trivalent [Cr(III)]. Chromium is widely used in various industries such as electroplating, leather tanning, chrome paint, dying, hardened steel, ceramic, and glass. Cr(VI) is produced almost exclusively by industrial processes; whereas, in nature chromium primarily exists in its trivalent form. Cr(III) is generally regarded as nontoxic due to its poor absorption. Cr(VI) is considered a pulmonary carcinogen and has tested positive in genotoxicity tests. Cr(VI) is considered a serious pollutant in many water streams due to its carcinogenic potential. In trace amounts, Cr(III) may be required for glucose and lipid metabolism.

Application

This kit is suitable for the detection of chromium in biological, environmental, food, and beverage samples.

Biochem/physiol Actions

In this kit, chromium is determined in a simple, one-step assay in which Cr(VI) forms a specific complex with a chromgenic dye, resulting in a colorimetric (480 nm) product, proportional to the chromium present. Cr(III) can be converted to Cr(VI) with nitric acid/hydrochloric acid, allowing the determination of Cr(III) and/or total chromium [Cr(III) +Cr(VI)].

  1. What is the storage condition for sample after oxidation with nitric acid & hydrochloric acid? Do I need to process the assay right after ?

    1. After oxidation with nitric acid and hydrochloric acid, testing should be performed promptly. Unused samples may be stored at -80 °C, but the method has not been validated. No stability data are available for samples stored after treatment.

  2. Can I use Sodium Hydroxide to neutralized the samples via oxidation with nitric acid /Hydrochloric acid? I will need to add a lot of 3% ammonia solution to make sample pH to 7

    1. This product has not been validated with Sodium Hydroxide for neutralization. Neutralization should be conducted with the recommended 3% ammonia solution for this kit, and any use of Sodium Hydroxide would require independent verification of compatibility.

  3. Regarding your MAK130 Chromium Assay Kit. When converting Cr3 to Cr6 in samples via oxidation with nitric acid. It states to "neutralize" the solution with 3% ammonia solution, it doesn't state the amount to use ie volume required for this?.

    1. The amount of ammonia solution required can very from one experiment to the next; it depends upon the initial acidity of the solution, which can vary. It is recommended to add the ammonia dropwise until neutralization is achieved.
