Biochem/physiol Actions

Two-chain glycoprotein hormone; the α-chain is identical to that of chorionic gonadotropin, TSH, and FSH, while biological specificity is attributed to the distinct β-chain. Induces ovulation, spermatogenesis, and synthesis of steroid sex hormones.
Two-chain glycoprotein hormone; the α-chain is identical to that of chorionic gonadotropin, TSH, and FSH, while biological specificity is attributed to the distinct β-chain. Induces ovulation, spermatogenesis, and synthesis of steroid sex hormones. Activity is mediated through a trans-membrane LH/chorionic gonadotropin receptor that is a member of the G-protein coupled receptor (GPCR) family.

Questions

1–6 of 6 Questions
  1. How do I store reconstituted LH? And for how long is LH stable after reconstitution?

    1 answer
    1. This product may be reconstituted at 1 mg/mL in 10 mM Sodium Phosphate, 150 mM Sodium Chloride containing 1 mg/ml BSA, 0.1% Sodium Azide, pH 7.4. Stock solutions may be stored in aliquots at -20°C. Avoid repeated freeze/thaw cycles. Frozen aliquots are expected to be stable up to 6 months.

  2. How can I determine the shelf life / expiration / retest date of this product?

    1 answer
    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.
      For more information, please refer to the Product Dating Information document: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/418/501/product-dating-information-06-25-mk.pdf

  3. How is shipping temperature determined? And how is it related to the product storage temperature?

    1 answer
    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. For more information, please refer to the Storage and Transport Conditions document: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/316/622/storage-transport-conditions-mk.pdf

  4. Could you provide the concentration of L6420 in nanograms per milliliter (ng/ml)?

    1 answer
    1. Product L6420 is a white powder that is lyophilized from a 50 mM ammonium bicarbonate solution. Since this product is not a solution, it does not have an associated concentration in ng/mL.

  5. What volumen of the buffer should be added to dilute the product?

    1 answer
    1. For a stock solution, hormones should be reconstituted to ≥1 mg/ml in the presence of a protease-free carrier (0.1-1% ovalbumin, BSA, or gelatin) and a suitable antimicrobial agent. The following general guidelines may be useful:

      1. Ovalbumin is generally used when assaying for gonadotropin;
      2. If BSA is used, it must be free of gonadotropins and proteases (either crystalline or RIA grade is generally satisfactory);
      3. Heat gelatin solutions to 60°C for maximum solubility (1% gelatin for ELISAs, 0.1% for solid phase assays);
      4. For the pituitary hormones hFSH, hLH, hPRL, and hTSH, the recommended dilution buffer is 10 mM Sodium Phosphate, 150 mM Sodium Chloride, 1 mg/ml BSA, 0.1% Sodium Azide, pH 7.4 is generally satisfactory for hFSH, hLH, hPRL, and hTSH.
      5. For hCG and hGH, the recommended dilution buffer is 100 mM Tris, 1 mg/ml BSA, 0.05% Sodium Azide, pH 8.0 is generally satisfactory for hCG and hGH.

  6. How to dilute the LH L6420

    1 answer
    1. It is recommended to dilute Product LH L6420 in a buffer consisting of 10 mM Sodium Phosphate, 150 mM Sodium Chloride, 1 mg/ml BSA, 0.1% Sodium Azide (optional), pH 7.4.
