Application

α-glucosidase is used for the determination of α-amylase and the synthesis of various 1′-O-sucrose and 1-O-fructose esters. It was also used in the measurement of glycosidase inhibition.[1]

Biochem/physiol Actions

α-glucosidase hydrolyzes carbohydrates by acting on 1,4-α linkages. Inhibition of α-glucosidase is a prominent target in the management of non-insulin-dependent diabetes mellitus[2].
Hydrolysis of terminal, non-reducing 1→4-linked D-glucose residues with release of D-glucose.

Packaging

Sold on basis of p-nitrophenyl α-D-glucoside units.

Protein determined by biuret.

Other Notes

One unit will liberate 1.0 μmole of D-glucose from p-nitrophenyl α-D-glucoside per min at pH 6.8 at 37 °C.

    1. The protein content and enzymatic activity are determined on a lot-to-lot basis. Each value is reported in the lot-specific Certificate of Analysis. The minimum protein content is 50% and the minimum enzymatic activity is 10 units per milligram protein. A review of recent lots indicates typical protein concentrations of >60% and activities of 15 - 30 units per milligram protein.     1. Literature indicates that the substrate concentration for using 4-Nitrophenyl α-D-glucopyranoside with this enzyme is typically in the saturation range of 1 mM or higher.

    1. The molecular weight of this enzyme has not been determined. The chain structure, number of sulfide bonds, and other parameters have not been characterized. However, a similar recombinant version, product G0660, lists a molecular weight of approximately 68.5 kDa. Other sources report the molecular weight of a-Glucosidase from Saccharomyces cerevisiae at approximately 63 kDa.
