Specificity
Clone 1H17 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects LIN28B. It targets an epitope within 22 amino acids from the C-terminal half.

KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal half of human LIN28B.
Application
Quality Control Testing
Evaluated by Western Blotting in K562 cell lysate.
Western Blotting Analysis): A 1:10,000 dilution of this antibody detected LIN28B in K562 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected LIN28B in HepG2 cell lysate.
Flow Cytometry Analysis: 1 µg from a representative lot detected LIN28B in one million K562 cells.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected LIN28B in HepG2 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected LIN28B in Human placenta tissue sections.

Target description
Protein lin-28 homolog B (UniProt: Q6ZN17; also known as LIN-28B) is encoded by the LIN28B (also known as CSDD2) gene (Gene ID: 389421) in human. LIN28B is an evolutionarily conserved RNA-binding protein that regulates mRNA translation and miRNA let-7 maturation in embryonic stem cells and developing tissues. Two isoforms of LIN-28B have been described that are produced by alternative splicing. Isoform 1 is detected in placenta and in poorly differentiated hepatocellular carcinoma cells. Isoform 2 is detected in fetal liver, non-tumor liver tissues, as well as well-differentiated tumor tissues. LIN-28B is predominantly nucleolar and its nucleolar localization signal resides in amino acids 239-250. It serves as a suppressor of microRNA (miRNA) biogenesis, including that of let-7 and possibly of miR107, miR-143 and miR-200c. It binds primary let-7 transcripts (pri-let-7), including pri-let-7g and pri-let-7a-1, and sequester them in the nucleolus, away from the microprocessor complex, hence preventing their processing into mature miRNA. In mammals, the LIN28B plays a critical role in multiple physiological processes, including skeletal myogenesis, neurogliogenesis, lymphopoiesis, germ-cell development, and glucose metabolism. LIN-28B levels ae up-regulated in triple-negative (ER-,PR-,HER2-) breast tumors, as well as in liver, ovarian, and thyroid carcinomas. It is shown to inhibit apoptosis through regulation of the AKT2/FOXO3A/BIM axis in ovarian cancer cells. When overexpressed, isoform 1 is reported to stimulates growth of the breast adenocarcinoma cell line MCF-7, whereas isoform 2 does not affect cell growth. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
