Biochem/physiol Actions

C11-BODIPY581/591 is an oxidation-sensitive fluorophore whose maximal ex/em wavelengths shift from 581/591 nm (red) to 500/510 (green) upon lipid peroxidation (LPO). The probe incorporates readily into lipid membranes and is useful for detecting and quantifying LPO of cell-free lipid preparations as well as in live cells by fluorometer (fluorimeter), fluorescence confocal microscopy (dual ex at 488 & 568 nm, detection by 530/30 nm and 590/30 nm bandpass filters) and flow cytometry (ex/em 484/510 for green, ex/em 581/610 for red signal).

Here, we identify iPLA2β as a critical regulator for p53-driven ferroptosis upon reactive oxygen species (ROS)-induced stress. The calcium-independent phospholipase iPLA2β is known to cleave acyl tails from the glycerol backbone of lipids and release oxidized fatty acids from phospholipids.
