General description

Carnitine is a metabolite that is synthesized from lysine and methionine. Carnitine is essential for fatty acid β-oxidation due to its role in fatty acid transport into the mitochondrial matrix via the carnitine/acylcarnitine shuttle. Carnitine exists in two stereoisomers but only the L-carnitine isomer is biologically active. Tissue levels of L-carnitine decrease with aging and decreased L-carnitine may contribute to age-related mitochondrial decline. The L-Carnitine Assay Kit is a simple convenient means of measuring free L-Carnitine in biological samples such as serum.

Application

Suitable for the measurement of free L-Carnitine in biological samples such as tissue, cells and serum
L-Carnitine Assay Kit has been used to determine the concentration of carnitine in serum samples.

Biochem/physiol Actions

L-carnitine concentration is determined by a coupled enzyme assay, which results in a colorimetric (570 nm)/ fluorometric (λex = 535/λem = 587 nm) product, proportional to the L-carnitine present. Typical sensitivities of detection for this kit are 10 μM (colorimetric) and 1 μM (fluorometric). The normal range for serum L-Carnitine is between 10-70 μM.

Questions

1–3 of 3 Questions
  1. for how long can plasma or serum samples be frozen?

    1 answer
    1. L-Carnitine in solution appears to be relatively stable when stored at –20 °C. This kit includes an L-Carnitine standard. When reconstituted, this standard may be stored at –20 °C for 3 months. Serum samples would presumably have similar stability at –20 °C. Serum products that are offered for culture or similar uses, which are stored at –20 °C, have a general shelf life of 3 - 4 years.

  3. Can these kits be used with serum samples from mouse?

    1 answer
    1. Yes, this kit can be used on mouse serum samples. We generally recommend that fresh serum samples be used. Frozen samples can probably be used. However, the successful use of frozen samples depends on the efficiency of freezing them and the amount of time they have been frozen. If previously frozen serum samples are used, you may want to check the performance of the kit with fresh vs. frozen samples.
