General description

Mast cell chymase (EC 3.4.21.39), a member of the granzyme subfamily of the S1 family of peptidases, cleaves C-terminal to Phenylalanine, Tyrosine, Tryptophan, and Leucine residues (in order of preference). Chymase is stored in Mast cell granules within the cell and is the major secreated Mast cell protease. Secreated Mast cell chymase is believed to be involved in extracellular matrix degradation, vasoactive peptide generation, and regulation of gland secreation.

Specificity

Reacts with human mast cell chymase.

Immunogen

Chymase purified from human skin.

Application

Anti-Chymase Antibody, clone B7 is an antibody against Chymase for use in DB, IH.
Dot blotting: 0.5-1.0 μg/mL.

Immunohistochemistry: 0.5-1.0 μg/mL. For optimal results, tissues or cytospin preparations should be fixed in Carnoy′s Fluid (60% ethanol, 30% chloroform, 10% glacial acetic acid). Not recommended for use on formaldehyde fixed tissue.

Not recommended for use on Western blots.

Optimal working dilutions must be determined by end user.

Purified by protein G affinity chromatography. Liquid in PBS with 20% glycerol, pH 7.4.

Storage and Stability

Maintain at 2-8°C in undiluted aliquots for up to 12 months from date of receipt. Antibody may also be stored at -20ºC in undiluted aliquots.
