General description

Heparin is mainly responsible for the delay in the coagulation of blood. It enhances the antithrombin-mediated inactivation of proteases in the coagulation pathway.[1]
Heparin sodium, a sulfated polysaccharide from the glycosaminoglycan family, plays a vital role in cell culture, cancer biology, and drug discovery research. Its impact on protein and cell interactions makes it valuable for studying molecular and cellular processes, tumor modeling, exploring binding interactions, and as a cell culture supplement to support cell viability.

Application

  • Heparin sodium salt from porcine intestinal mucosa has been used in sLRP (soluble circulating low density lipoprotein receptor-related protein-1) purification from human plasma.[2]
  • It has been used as a supplement in M199 medium for culturing of human umbilical vein endothelial cells.[3]
  • It has been used as an anticoagulant agent during isolation of blood.[4]

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   4. please, could you explain me if this H3149 is one of 'high molecolar weight heparin'? thanks

    1. This product is a cell culture tested porcine heparin with a molecular weight range of 15-19 kDa. This material is considered a "high molecular weight heparin." According to the literaure, low molecular weight heparins are those with a molecular wieght of less that 8000 Da. See the link below to review a publication that may be helpful:

  5. Can this material be diluted to 10,000 U/mL and stored at 2C to 8C? Do you have stability data at this temperature for lower concentrations?

    1. Yes, this product remains biologically active in dilute solutions. Stock solutions prepared at 50 mg/mL in water are stable for up to 2 years at 2 - 8°C, when filter sterilized. According to literature, heparin diluted in physiological saline at a concentration of 10 to 100 units/mL is stable for at least 35 days when stored at 2 - 8°C.     1. The heparin sodium salt (H3149, CAS 9041-08-1) is unfractionated. This classification is reflected in its molecular weight, which ranges approximately from 9,000 to 21,000, with the majority falling between 17,000-19,000. As confirmed by our product and as indicated in the USP monograph, our heparin is unfractionated. This differs from fractionated Heparin, also known as Low Molecular Weight Heparin, which has a much lower molecular weight. According to the DBR, 3rd ed., p. 342, the molecular weight of our heparin is approximately 16,000-17,000.

  7. Can you recommend a suitable heparin product for mouse heart perfusion?

    1. Our available heparin products have not undergone specific testing for mouse heart perfusion. However, you may find H3149 of interest as it has been cited in the following publication for this intended application: "Isolation and Primary Culture of Mouse Aortic Endothelial Cells" published in J Vis Exp. 2016; (118): 52965 (PMID: 28060318).

  8. Is it a different substance from heparan sulfate?

  9. Can I dissolve the heparin in filtered (0.22 um) nuclease free water at a concentration of 50mg/ml? Does it correspond to 5000 IU/ml? Then, I can use 15-30 IU/ ml of blood to avoid its coagulation, is it correct?

    1. As mentioned on the Product Detail Page, this product is soluble at 50 mg/mL in water. The actual activity is reported on the lot-specific Certificate of Analysis. This value should be used in determining the activity of the prepared solution. The activity specification for this product is >/=180 USP units/mg. For example, assuming an activity value of 200 units/mg, a prepared solution of 50 mg/ml will have an activity of 5000 units/ml. Yes, typically, 15 - 30 units/ml is suitable for use as an anticoagulant.

  10. Can we store the solution of Heparin sodium from intestinal mucosa of pig in RNase free water at -25 C?

    1. Solutions can be stored at 2-8 °C for up to 2 years, if sterile filtered through a 0.22 micron filter. The freeze/thaw process may cause degradation of the polysaccharide.
