General description

Protocolos

If an antibody must be purified from a small sample volume (0.5-10 mL), then the Amicon® Pro Purification System is recommended.

Detergents are frequently used to solubilize proteins and nucleic acids during purification, but the presence of detergents may interfere with downstream analyses.

Questions

1–7 of 7 Questions
  1. What is the margin of error for the MWCO? I read somewhere the RECOMMENDATION is to use a MWCO 2-3x smaller than your target. But I was wondering what’s the standard deviation from 3 kDa?

    1 answer
    1. For most membranes, the nominal molecular weight limit, also referred to as the molecular weight cut-off, is determined experimentally under a standard set of operating conditions. These analyses typically employ purified globular proteins to serve as markers or indicators of the retention characteristics of an ultrafiltration membrane. An ultrafiltration membrane with a stated nominal molecular weight limit should retain (reject) at least 90% of a globular solute of that molecular weight in daltons. However, for a wider safety margin, the selected cut-off should be well below the molecular weight of the solute to be retained. It is recommended to use a cut-off 2-3 times smaller than the molecular weight of the protein to be concentrated.

  2. Is this product compatible with organic solvents, such as DMSO or DMF?

    1 answer
    1. The Amicon Ultra products are suitable for use with samples containing up to 10% DMSO and 10% DMF. Exposure to 100% DMSO is not recommended. Unfortunately, the compatibility of these devices with 100% DMF has not been determined.

  3. Can I order the filter only, coming with no tube part?

    1 answer
    1. Unfortunately, no, this product is not available as a stand alone item. The tubes are available separately under the product UFC50VL96.

  4. Is the 0.5 ml 3KDa Amicon device (UFC5003) compatible with a solution containing 5% SDS (Sodium Dodecyl Sulfate)?

    1 answer
    1. The Amicon® Ultra centrifugal devices are designed for processing biological fluids and aqueous solutions. Amicon® Ultra Filter Devices are compatible with concentrations of Sodium dodecyl sulfate (SDS)of 0.1% or less. Prior to use, it is important to verify the chemical compatibility of the sample with the device.

  5. Hi, Do these filters come with expiry dates? If so where would I be able to find them on the product?

    1 answer
    1. This product is not assigned an expiration date or recommended retest date. Products with no expiration date or recommended retest date should be routinely inspected by customers to ensure they perform as expected. These products are also subject to a one year warranty from the date of shipment. 6. How long should I centrifugate at 14.000 g and how long for the reverse spin at 1000 g?

    1 answer
    1. Typical concentration time for the 3kD membrane device is approximately 30 minutes. However, spin times may need to be adjusted to either a shorter or longer time depending on the sample starting volume, initial concentration, composition, and desired minimum volume. The reverse spin is 1000g for 2 minutes. Please review the product user guide at the link below:

  7. Hi, how many g's can be used for the ultracell-3K #UFC500396 product?

    1 answer
    1. The recommended speed with a 35 degree fixed angle rotor is 14,000 g with a reverse spin of 1000 g. This information can be found at this link: https://www.sigmaaldrich.com/products/protein-biology/protein-sample-prep/amicon-centrifugals#ultracentrifugal
