Descripción general

"The SARS-CoV-2 coronavirus is the pathogenic cause of the human disease COVID-19. The virus particle consists of RNA encapsulated by the nucleocapsid (N) protein which is enveloped by a membrane (M) protein. The corona-like spike proteins surrounding this inner core consist of subunits S1, S2 and the receptor binding domain which binds to the human cellular ACE-2 receptor. Upon binding, the virus enters the cell, takes over the cellular machinery, reproduces, and the viral contagion continues. Viral structural proteins offer antigenic potential for an immune response. Upon viral infection the humoral immune system responds first by producing the immunoglobulin IgM, followed later by IgG which can convey long lasting immunity. IgA, a mucosal immune response, is also found in blood.By testing for IgM, IgG, and IgA in human serum and plasma, researchers may identify those who have been exposed to the SARS-CoV-2 virus and mounted some level of immune response. Researchers may further understand the immune response over the course of infection and recovery from COVID-19."MILLIPLEX® SARS-CoV-2 Antigen Panel 1 IgG is a 4-plex kit to be used for the simultaneous analysis of human serum or plasma samples for IgG antibodies which recognize any or all of the following analytes: SARS-CoV-2 Spike S1, SARS-CoV-2 Spike S2, SARS-Cov-2 RBD, Sars-CoV-2 N.

Aplicación

The SARS-CoV-2 Antigen Panel 1 IgG is a bead-based multiplex assay based on the Luminex® xMAP technology which enables the simultaneous analysis of human samples for IgG antibodies which recognize any of the following analytes: SARS-CoV-2 Spike S1, SARS-CoV-2 Spike S2, SARS-Cov-2 RBD, Sars-CoV-2 N.This is assay yields results in Median Fluorescence Intensity (MFI).Four assay control beads are included in the base kit.This is a one day assay.This assay requires 25 μL of 1:100 diluted plasma or serum.Analytes available:SARS-CoV-2 Nucleocapsid Protein (N);SARS-CoV-2 Receptor Binding Domain (RBD);SARS-CoV-2 Spike Protein Subunit 1 (S1);SARS-CoV-2 Spike Protein Subunit 2 (S2)

  1. Have the Milliplex Milliplex Human SARS-CoV-2 assays been tested on species other than humans?

    1. The Human SARS-CoV-2 kits were not tested with cross-species samples, so it is unknown whether they will detect samples from other species. There are no references available for using the kits with other species.

  2. Could you provide information on the subtypes detected by the SARS-CoV-2 IgG panel?

    1. The antibody was generated in goats and is probably designed to detect total IgG. It does not target a specific IgG subunit.

  3. We are running the SARS-CoV-2 IgG, IgA, and IgM kits for the first time on the MagPix. What analysis settings should be used since the kit does not include a standard? Are there any units that should be entered?

    1. For Qualitative Assays, the Analysis Type is "None" in xPonent. There are no units to be entered. Median values are used for the data comparison between treatment groups.

  4. What is the method for determining positive samples?

    1 answer
    1. To determine positive samples, it is necessary to run a number of negative control, pre-COVID, samples in the assay and obtain an average MFI for each antigen to compare to the unknown samples. It's important to note that the positive and negative controls do not play a role in determining whether customer samples are positive or negative for COVID.
