General description

Amicon® Ultra 15 mL centrifugal devices with Ultracel® 3,000 Dalton molecular weight cutoff filters provide fast ultrafiltration, and reliable retention of macromolecules. For example, Cytochrome C (12.4 kDa) at 0.25 mg/ml initial concentration is retained at >90% .

The vertical design and available membrane surface area provide fast sample processing, high sample recovery (typically greater than 90% of dilute starting solution), and the capability for 80-fold concentration. Solute polarization and subsequent fouling of the membrane are minimized by the vertical design, and a physical deadstop in the filter device prevents spinning to dryness and potential sample loss.

The concentrate is collected from the filter device sample reservoir using a pipetter, while the ultrafiltrate is collected in the provided centrifuge tube. The device can be spun in a swinging bucket or fixed angle rotor. With their vertical membrane design and deadstop, Amicon® Ultra 15 mL filters deliver the highest recovery with short spin times.

Application

  • Ultrafiltration with 3,000 Dalton molecular weight cutoff Ultracel® membrane.
  • Concentration of biological samples containing antigens, antibodies, enzymes, nucleic acids, or microorganisms.
  • Purification of macromolecular components found in tissue culture extracts or cell lysates and protein removal prior to HPLS.
  • Concentration of dilute or pre-purified proteins from column eluents.
  • Desalting, buffer exchange and protein dialysis.

Features and Benefits

  • Ultracel® regenerated cellulose membrane with 3 kDa NMWCO delivers retentate recovery of greater than 90%.
  • Vertical membrane reduces concentration polarization for ultra-fast spin times (as fast as 10–15 minutes).
  • Heat-sealed membrane minimizes downstream extractables.
  • 100% integrity tested for reliable performance.
  • Convenient sample monitoring with translucent housing and volume gradations.
  • Direct pipettor sample access eliminates processing step to recover concentrate.
  • High concentration factors of 80–100X.

Questions

1–6 of 6 Questions
  1. Hello, I did not get if all the proteins will remain in the filter and if I can recover them from the filter (product: UFC9003 Millipore Amicon® Ultra Centrifugal Filter, 3 kDa MWCO). What will exactly remain in the liquid part in the bottom? Many thanks

    1 answer
    1. These centrifugal devices separate constituents of a given sample by molecular weight. This product has a molecular weight cut-off (MWCO) of 3000 Daltons. Components of the sample with a molecular weight greater that the designated 3 kDa will remain in the retained portion following centrifugation, while components of less that 3 kDa will filter through. See the image below to review this 2 part device.

  2. Hi, I am writing to ask if you have centrifugal filters with hydrophobic membranes. I am looking to test them for oil/water separation downstream to a fermentation process for food applications.

    1 answer
    1. The ultrafiltration membranes used in Amicon devices are all hydrophilic. There are no hydrophobic options.

  3. Even sanitizing the devices with 70% ethanol, how many times may I use one tube? I mean, How many times can be used it?

    1 answer
    1. The unit is for one-time use only.

      Helpful?

  4. How many tubes are inside the pack? For example, there is a pack size of 8 pkg. What does it mean? Does it mean 8 tubes or 8 packs of tubes? Does it mean that there are 8 tubes in the pack? Can you explain more, please?

    1 answer
    1. The package quantity for each item is as follows:
      UFC900308 - 8 tubes
      UFC900324 - 24 tubes
      UFC900396 - 96 tubes

      Helpful?

  5. How much g (rcf) force can be applied during the spin?

    1 answer
    1. Amicon® Ultra-15 Centrifugal Filter Devices can be spun at 4,000 x g maximum for approximately 15-60 minutes when using a swinging-bucket rotor, or at 5,000 x g maximum for approximately 15-60 minutes when using a fixed-angle rotor.

      Helpful?

  6. Is autoclave sterilization possible?

    1 answer
    1. The Amicon Ultra devices are manufactured under very thorough and controlled cleaning conditions, however they are not sterile and cannot be sterilized by UV or autoclaving. However, it is possible to sanitize the devices with 70% ethanol, followed by several rinses with sterile water or culture medium.
