The pIEx™-4 DNA contains C-terminal S•Tag and His•Tag coding sequences, whereas the pIEx™-6 DNA contains an amino-terminal His•Tag coding sequence and an optional S•Tag coding sequence at the distal end of the multiple cloning site (MCS).
Additionally, the pIEx™-6 DNA features a PshA I cloning site that overlaps the cleavage site for the enterokinase (EK) protease, enabling the removal of all amino-terminal vector-encoded sequences by EK digestion, which is not the case for pIEx™-4 DNA.
Furthermore, neither pIEx™-4 DNA nor pIEx™-6 DNA offers the option of using a signal sequence to secrete the protein of interest into the surrounding medium.
