General description

The pIEx vectors are designed for cloning and high-level expression of proteins in transiently transfected Spodoptera-derived insect cells. Transcription is driven by the AcNPV-derived hr5 enhancer and immediate early promoter, IE1.

pIEx-4 contains C-terminal S•Tag and His•Tag coding sequences. For sequencing or PCR of recombinants in pIEx-4, refer to the table below to choose the appropriate primers.

  1. What are the significant differences between vector pIEx-4 and pIEx-6? The aim is to express a membrane-specific protein without needing any signal peptidase at the 5' end.

    1. The pIEx™-4 DNA contains C-terminal S•Tag and His•Tag coding sequences, whereas the pIEx™-6 DNA contains an amino-terminal His•Tag coding sequence and an optional S•Tag coding sequence at the distal end of the multiple cloning site (MCS).

      Additionally, the pIEx™-6 DNA features a PshA I cloning site that overlaps the cleavage site for the enterokinase (EK) protease, enabling the removal of all amino-terminal vector-encoded sequences by EK digestion, which is not the case for pIEx™-4 DNA.

      Furthermore, neither pIEx™-4 DNA nor pIEx™-6 DNA offers the option of using a signal sequence to secrete the protein of interest into the surrounding medium.
