General description

Large envelope protein (UniProt: A7XR30; also known as L glycoprotein, L-HBsAg, LHB, Large S protein, Large surface protein, Major surface antigen) is encoded by the S (also known as Large S, P, preS1, preS2) gene (Gene ID: 944569) in Hepatitis B virus (HBV). HBV is a small enveloped virus of the Hepadnaviridae family with a partially double-stranded DNA genome approximately 3.2 kb. Hepatitis B surface antigen is a distinctive serological marker of acute or chronic hepatitis B infection and is the first antigen to appear following infection with hepatitis B virus. HBV contains three viral envelope/surface proteins - large surface antigen, middle surface antigen, and small surface antigen. They are expressed from a single open reading frame. The large envelope protein is synthesized with the pre-S region at the cytosolic side of the endoplasmic reticulum and is not N-glycosylated. It is shown to exist in two topological conformations, one which is termed ′external′ or Le-HBsAg and the other ′internal′ or Li-HBsAg. In its external conformation the protein attaches the virus to cell receptors and thereby initiating infection. This attachment induces virion internalization predominantly through caveolin-mediated endocytosis. The large envelope protein also assures fusion between virion membrane and endosomal membrane. In its internal conformation the protein plays a role in virion morphogenesis and mediates the contact with the nucleocapsid like a matrix protein. (Ref.: Wu, CC., et al. (2018). World J.
