一般描述
双缩脲和 Lowry 程序用于蛋白质测定。前者广泛用于临床检测。后者尽管更灵敏，却只用于研究工作，受限于组合试剂的低稳定性、颜色的不可再现性（特别是在低蛋白浓度下）以及和蛋白质浓度的非线性显色反应。Ohnishi 和 Barr 改良了用于 Lowry 测定法的双缩脲试剂，从而简化了流程，同时提高了组合试剂的稳定性。
应用
全蛋白试剂盒（ Micro-Lowry, Onishi & Barr Modification ）已用于测定以下物质的总可溶蛋白质浓度:
- 麦醇溶蛋白的透析液/提取物
- 面粉（燕麦 和 硬粒小麦）和种子（藜麦, 芡欧鼠尾草）的提取物
- 储存的大鼠肝微粒体部分样品
原理
比色法，终点。 化验。Ohnishi 和 Barr′s 对微 Lowry 方法的改进。根据实验步骤，稀释的缩二脲试剂与肽键反应生成紫蓝色复合物，通过加入苯酚试剂增强其颜色。在 550-750 nm 处读取的吸光度用于确定标准曲线的结果。
