GlycoProfile IV 试剂盒使用了去糖基化系统中的三氟甲磺酸(TFMS),可完全去除N- 和O-连接聚糖,同时保护蛋白质或多肽结构。TFMS作用下的糖蛋白水解使得蛋白质分解量达到最小,同时消除了聚糖的释放。反应条件经过优化,无需清除剂即可在30分钟内使简单或低分子量糖蛋白脱糖基。对于高分子量或复杂的非哺乳动物糖蛋白,以无水茴香醚为清除剂可以得到高蛋白质产率。

應用

GlycoProfile IV 化学去糖基化试剂盒已被用于人αL-艾杜糖苷酸酶和来自棉铃虫肠道的围食膜基质(peritrophic matrix )的去糖基。它已被用于评估植物重组人白细胞介素13(rhIL-13)的糖基化。
