一般描述

ONPG（2-硝基苯-β-D-半乳糖苷）是β-半乳糖苷酶的比色底物。

应用

2-硝基苯-β-D-半乳糖苷是一种酶底物，可用于检测lacZ活性，从而检测是否存在β-半乳糖苷酶。

生化/生理作用

β-半乳糖苷酶可将乳糖分解为半乳糖和葡萄糖。β-半乳糖苷酶不是乳糖特异性的，并且可以水解简单的半乳糖苷。2-硝基苯-β-D-半乳糖苷水解，导致半乳糖和黄色显色化合物的释放。该测试底物不依赖于诱导或组成型通透酶进入细胞，因此反应迅速并且在24小时内发生。

原理

β-半乳糖苷酶可将乳糖分解为半乳糖和葡萄糖。β-半乳糖苷酶不是乳糖特异性的，并且可以水解简单的半乳糖苷。2-硝基苯-β-D-半乳糖苷水解，导致半乳糖和黄色显色化合物的释放。该测试底物不依赖于诱导或组成型通透酶进入细胞，因此反应迅速并且在24小时内发生。

重悬

可使用分子生物学级水制备浓度为3 mg/ml的储备溶液。 也可使用100 mM磷酸钠缓冲液（pH 7.3）制备约20.5 mg/ml的储备溶液。可能需要稍微加热，才能完全溶解本品。
