一般描述

MES水合物(或称4-吗啉乙磺酸水合物)是一种两性离子吗啉缓冲液,它是一种众所周知的“Good′s”缓冲液,适用于生物学和生物化学应用。在5.5-6.7 pH范围内的良好缓冲能力使其适合各种研究应用。特别值得一提的是,MES缓冲液最大程度减少了与环境和生物学研究中使用的常见金属的相互作用,从而防止了对实验结果的干扰。

MES缓冲液具有良好的水溶性并且不溶于脂质,它不能够渗透细胞膜,这一点确保了它在细胞环境中的功效。它的应用包括维持细胞培养基、蛋白质基缓冲液和电泳缓冲液的稳定性。MES缓冲液可用于纯化抗体、肽、蛋白质、血液成分和生长因子。MES 缓冲液可作为甲次砷酸盐和非两性离子缓冲液(如柠檬酸盐和苹果酸盐)等有毒试剂的更良好的替代品。在各种实验环境中,MES 缓冲液可用于调节生长培养基的pH值,稳定酶溶液,并可作为PAGE电泳缓冲液的成分。

制备说明

可通过NaOH滴定至所需的pH值,制备使用MES的缓冲液。也可将MES和MES钠盐的储备溶液混合,达到所需的pH。目前已公布了使用储备溶液制备指定 pH 缓冲液的标准混比表[5]。不建议在pH 7.4缓冲条件下使用MES;应考虑其他缓冲液。[6]

储存及稳定性

溶液可在2-8℃下稳定保存数月。通过0.2uM过滤器过滤灭菌。不建议对任何磺酸缓冲液进行高压蒸汽灭菌。如果缓冲液必须不含核酸酶,请先处理水,然后在高压蒸汽灭菌后添加缓冲液。MES溶液高压蒸汽灭菌后,会变成黄色(尽管pH值并未明显变化)。黄色分解产物具体成分未知。

  1. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.
