原則
当 Blue-White Select ™ 筛查试剂被 β-半乳糖苷酶裂解时，所得产物会形成蓝色沉淀。存在 Lac + 菌落的情况下生长 Select ™ 筛选试剂保持白色，便于区分 lac + 和 lac-菌落。
该技术是基于载体的，如携带编码β-半乳糖苷酶 α-片段的 β-半乳糖苷酶基因的 pUC 和 M13mp 系列。开发这些载体需要使用携带互补基因片段的细菌菌株，以允许组装活性复合物，从而形成蓝色菌落。将 DNA 片段插入载体导致 ß-半乳糖苷酶基因断裂 ′多克隆位点导致功能 β-半乳糖苷酶活性；这些菌落保持白色，便于区分 lac + 和 lac-菌落。
