Allgemeine Beschreibung

Der Antikörper erkennt ein Epitop am C-Terminus von Aktin, der in allen Aktin-Isoformen konserviert ist. Er markiert gezielt Aktin in einer Vielzahl von Geweben und Spezies. Das vom Antikörper erkannte Epitop ist resistent gegenüber Formalinfixierung und Paraffineinbettung. Zink-Formalin, B5, Ethanol, Methacarn und Brunnel- oder Bouin-Lösung können ebenfalls als Fixiermittel verwendet werden.
Monoclonal Anti-Actin (mouse IgG2a isotype) is derived from the AC-40 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice. It has one N-terminal region which appears to be a major antigenic region and may be involved in the interaction of actin with other proteins such as myosin. Actin isoforms show 50-60% homology in their 18 N-terminal residues.

Anwendung

Als primärer Antikörper für die Western-Blot-Analyse von MDCK-Zelllysaten wurde ein monoklonaler Anti-Aktin-Antikörper verwendet.
Monoklonaler Anti-Aktin-Antikörper wurde für die Western-Blot-Analyse von Cos-7-Zelllysaten verwendet, um eine gleichmäßige Beladung von Proteinen zu garantieren.
Actin–FITC antibody is suitable for direct immunofluorescence at a dilution of 1:75, using cultured human or chicken fibroblasts.

Biochem./physiol. Wirkung

Actin, a conserved eukaryotic protein, is the major cytoskeletal component which actively participates in cell motility. It is associated with myriad of cellular processes including locomotion, secretion, cytoplasmic streaming, phagocytosis and cytokinesis. In mammals and birds, actin is expressed as six different isoforms. It has one N-terminal region which appears to be a major antigenic region, and may be involved in the interaction of actin with other proteins such as myosin. Actin isoforms show 50-60% homology in their 18 N-terminal residues.

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 1% bovine serum albumin and 15 mM sodium azide.
