To measure hyaluronidase activity, a turbidimetric determination assay is used at 600 nm. One unit of hyaluronidase activity will cause a change in absorbance of 0.330 per minute at pH 5.35 at 37 °C.

Is there any information available regarding whether hyaluronidase impacts body fluid crystals?

Hyaluronidase is not expected to impact crystals. It is specific to hyaluronan or Hyaluronic acid and may be used with breast and possibly other tissues. However, crystals are typically inorganic salts, with body fluid crystals typically being pyrophosphate. Therefore, hyaluronidase should have no effect upon pyrophosphate crystals.

Is it acceptable to use H3506 SLCH0779 directly in synovial fluid without dilution?

It is generally not a standard practice to simply sprinkle hyaluronidase (such as H3506 SLCH0779) directly into synovial fluid. For a more formal and generally recognized method, a 0.05% solution in phosphate buffer is recommended, with one drop added per milliliter of fluid. This allows for the clarification of synovial fluids to make cell counts possible. Therefore, directly using H3506 SLCH0779 in synovial fluid without dilution may not be the most common practice.

Can I dissolve H3506 in DMEM/F12 media or PBS before use for tissue dissociation?

It is recommended to dissolve this product at 1 mg/mL in 0.02 M phosphate buffer, pH 7, containing 77 mM sodium chloride and 0.01% BSA. Solubilizing directly into the medium has not been tested, but may be suitable. It is important to include BSA to ensure stability.
