ICCB-19 is a cell penetrant apoptosis inhibitor that activates autophagy leading to restoration of cellular homeostasis in cells with accumulated mutant tau, α-synuclein, or huntingtin. ICCB-19 potently and selectively inhibits TRADD. It binds to the N-terminal TRAF2-binding domain of TRADD (TRADD-N). Apparently ICCB-19 releases cIAP1/2 or TRAF2 from TRADD thus promoting the K63-linked ubiquitination of beclin 1.
