Nebulin is a 700kDa protein which is expressed in skeletal muscles. It possesses 35 amino acid repeats and a C terminal serine-rich and SH3 domains.
Nebulin is also localized to cardiac muscle. Monoclonal Anti-Nebulin (mouse IgG1 isotype) is derived from the hybridoma produced by the fusion of mouse myeloma cells and splenocytes from an immunized mouse.
Specificity
Localizes nebulin in skeletal muscle of a wide variety of species from cold-blooded vertebrates to man. Does not crossreact with smooth or cardiac muscle, non-muscle tissue and cultured cells. Localizes nebulin within the I band of skeletal muscle sections by indirect immunofluorescence.
Immunogen
chicken breast nebulin.
Application
Monoclonal Anti-Nebulin antibody produced in mouse has been used in:
immunohistochemistry
fluorescent immunostaining
western blotting
Biochem/physiol Actions
Nebulin associates with the Z-line and thin actin filament in a sarcomere. It may function as a regulator of sarcomere thin filament length. The protein also acts as a structural link between sarcomeres. Nebulin interacts with the capping protein CapZ and has a role in controlling the localization of actin filaments.

cDNA clones encoding mouse skeletal muscle nebulin were expressed in Escherichia coli as thioredoxin fusion proteins and purified in the presence of 6 M urea. These fragments, called 7a and 8c, contain 28 and 19 of the weakly repeating approximately
