General description

Formate:NAD⁺ oxidoreductase
Formate dehydrogenase is part of the D-specific 2-hydroxy acid superfamily of dehydrogenases. It is a dimer of two identical subunits. The enzyme has a cysteine residue which is essential for its activity or structural integrity.

Application

Most widely used in cofactor recycling systems for NADH.

Biochem/physiol Actions

Substrate Specificity and K_m:
Formate dehydrogenase reacts only with formate (K_m = 13 mM at 30 °C and pH 7.5; 11 mM at 25 °C and pH 7.5) and NAD (K_m 0.09 mM; 30 °C; pH 7.5). It does not react with acetate, oxalate, lactate, succinate, propionate or ascorbate, nor will the enzyme reduce NADP.

One unit (U) formate dehydrogenase will oxidize 1 μmol of formic acid to CO₂ in 1 minute at +25 °C and pH 7.6. The above assay produces 1 mmol of NADH per mmol of formate oxidized.
