Diquat metabolite dipyridone is a common degradation product of diquat. Diquat is a nonselective, defoliant, preharvest, and non-residual bipyridyl herbicide and crop desiccant, normally used as the dibromide salt, to control broad-leaved weeds and grasses. Its activity is based on the liberation of the superoxide anion radical and, subsequently, hydrogen peroxide, leading to tissue destruction by oxidative stress. The herbicidal activity of diquat is more pronounced in daylight, aerobic conditions, high humidity, and temperature.

As per Regulation (EC) No 1107/2009, repealing the directive 91/414/ECC, diquat is not approved for use in the European Union. The maximum residue limit (MRL) for diquat presence in the crops varies between 0.01 mg/kg in most of the commodities to 5 mg/kg for linseed in accordance with Regulation (EU) 2016/1002.

Application

The certified reference material (CRM) is intended to be used as a calibrant for chromatography and other analytical techniques.
The Diquat metabolite dipyridone CRM may also find the following uses:
  • Determination of paraquat, diquat, and two metabolites of diquat (diquat-monopyridone and diquat-dipyridone) in biological materials using high-performance liquid chromatography combined with UV and fluorescence detectors
  • Detection of two metabolites of diquat (diquat-monopyridone and diquat-dipyridone) in urine and serum of poisoned patients after ingestion of a combined herbicide of paraquat and diquat
  • Simultaneous quantitation of diquat and its two metabolites in serum and urine by ion-paired high-performance liquid chromatography
  • To develop a fast ultraperformance liquid chromatography (UPLC)-MS/MS method for the determination of diquat and its two primary metabolites in rat plasma

The determination of paraquat, diquat and two metabolites of diquat in biological materials was developed using high-performance liquid chromatography combined with UV and fluorescence detectors. Paraquat, diquat and internal standards (ethyl viologen and o-acetamidophenol) were detected by the UV detector.
