Specificity
Clone 1E15 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Ubiquitin carboxyl-terminal hydrolase 11 (USP11). It targets an epitope within 25 amino acids from the N-terminal half.
Immunogen
KLH-conjugated linear peptide corresponding to 25 amino acids from the N-terminal half of human Ubiquitin carboxyl-terminal hydrolase 11 (USP11).
Application
Quality Control Testing
Evaluated by Western Blotting in HepG2 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected USP11 in HepG2 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected USP11 in HeLa cell lysate.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected USP11 in Human cerebral cortex tissue sections .

Target description
Ubiquitin carboxyl-terminal hydrolase 11 (UniProt: P51784; also known as EC:3.4.19.12, Deubiquitinating enzyme 11, Ubiquitin thioesterase 11, Ubiquitin-specific-processing protease 11) is encoded by the USP11 (also known as UHX1) gene (Gene ID: 8237) in human. USP11 is a monomeric protein of the peptidase C19 family that can remove conjugated ubiquitin from target proteins and polyubiquitin chains. It is mainly localized in the nucleus with only negligible part present in the cytoplasm. It inhibits the degradation of target proteins by the proteasome. It is involved in the regulation of multiple processes in cells, including cell cycle, DNA repair, tumor development, and signaling. It also acts as a chromatin regulator via its association with the Polycomb group (PcG) multiprotein PRC1-like complex; may ACT® by deubiquitinating components of the PRC1-like complex. It is reported to deubiquitinate and stabilize NRF2, thereby regulating cell proliferation and ferroptotic cell death. Depletion of USP11 is known to suppression of cell proliferation and induce of ferroptotic cell death due to ROS-mediated stress that can be largely abrogated by overexpression of NRF2. Its USP domain is localized in amino acids 309-930. USP11 cleaves preferentially lysine 6- and lysine 63-linked ubiquitin chains and displays lower activity with lysine 11- and lysine 33-linked ubiquitin chains, and extremely low activity with lysine 27-, 29-, and 48-linked ubiquitin chains. Dysregulation of USP11 is observed in several human tumors, including melanoma, glioma, colorectal cancer, and cervical cancer. It is reported to promote tumorigenesis by stabilizing cellular inhibitor of apoptosis protein 2 in colon cancer and melanoma, however, it can also suppress tumorigenesis by stabilizing PTEN homolog in prostate cancer. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
