General description

Hygromycin B is an aminoglycoside antibiotic, inhibits the growth of prokaryotic and eukaryotic microorganisms and mammalian cells. Inhibits protein synthesis by interfering with the translocation of the 70S ribosome and causes misreading of mRNA. Resistance to hygromycin B is conferred by a gene whose product, hygromycin B phospho-transferase that phosphorylates and inactivates the drug. Hygromycin B has been used to select mutants carrying the hygromycin B resistance gene in a wide variety of cells including bacterial, protozoal, yeast, fungal, plant, and mammalian cells. Typical concentrations used for mammalian cell selection are between 0.05 to 1 mg/ml. The appropriate concentration for use must be determined empirically for each cell type.

Metabolism plays a critical role in direct regulation of a variety of cellular activities via metabolic enzymes and metabolites. Here, we demonstrate that phosphofructokinase 1 platelet isoform (PFKP), which catalyzes a rate-limiting reaction in glycolysis, promotes EGFR activation-induced nuclear translocation
