General description

Histone H4 (UniProt P02309) is encoded by the HHF1 (ORF YBR0122; Gene ID 852294) and HHF2 (ORF N2752; Gene ID 855701) genes in yeast species. Histone H4 is one of the 5 main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N terminal tail, H4 is involved with the structure of the nucleosomes of the ′beads on a string′ structure. Acetylation of histone H4 occurs at several different lysine positions in the histone tail and is performed by a family of enzymes known as Histone Acetyl Transferases (HATs).

Specificity

Broad species cross-reactivity is expected.
H4K16ac specificity of the unconjugated antibody (Cat. No. 07-329) has been confirmed using the Absurance Histone H3 Antibody Specificity Array (Cat. No. 16-667) and the Absurance Histone H2A, H2B, H4 Antibody Specificity Array (Cat. No. 16-665).

Immunogen

Epitope: H4K16ac
Ovalbumin-conjugated, synthetic peptide (…GAAcKRH …) in which AcK corresponds to residue 16 of yeast histone H4.

Application

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected acetyl-Histone H4 (Lys16) in A431 and NIH/3T3 cells.
The unconjugated antibody (Cat. No. 07-329) is shown to be suitable also for ChIP, ChIP-seq, Western blotting, multiplex assay, dot blot applications.
This Anti-acetyl-Histone H4 (Lys16) Antibody, AlexaFluor 488 is validated for use in Immunocytochemistry for the detection of acetyl-Histone H4 (Lys16).

Quality

Evaluated by Immunocytochemistry in HeLa cells.

Immunocytochemistry Analysis: A 1:100 dilution of this antibody detected acetyl-Histone H4 (Lys16) in HeLa cells.

Self-renewal and differentiation of hematopoietic stem cells (HSCs) are orchestrated by the combinatorial action of transcription factors and epigenetic regulators. Here, we have explored the mechanism by which histone H4 lysine 16 acetyltransferase MOF regulates erythropoiesis. Single-cell RNA sequencing and
