C18カラムでの方法をDiscovery® HS F5 HPLCカラムに移行するためのガイドライン:
塩基性物質は通常C18カラムよりもHS F5カラムのほうが長く保持されます。C18カラムでの分離における有機溶媒の濃度を5~10%上昇させると、通常HS F5カラムで類似した保持が再現されます。他の化合物での結果は大きく異なります。しかし、2.5未満のlog Po/w値の溶質はC18と比べてHS F5 では通常長く保持されます。差の程度は溶質に大きく依存しています。
Discovery® HS F5の結合相は、C18カラムとは極めて異なった逆相分離を提供します。しかし、化合物は通常同じ保持時間枠内に溶出されるので、多くの場合C18カラムでの方法を容易に移行できます。
