Biochem/physiol Actions

細胞死関連プロテインキナーゼ1(DAPK1)は、γ-インターフェロンで誘導されるアポトーシスに対する正のメディエーターです。DAPKの活性化はSer-308の脱リン酸化とそれに続くカルシウム/カルモジュリンとの結合を通じて起こります。DAPKは腫瘍壊死因子やセラミドに応答して急速に脱リン酸化され、プロテアソーム活性による分解を受けます。また、DAPKの発現低下に伴ってカスパーゼ活性の上昇や細胞のアポトーシスが生じています。DAPKによるアポトーシス制御の活性は、リン酸化状態とタンパク質の安定性の両方によってコントロールされることが複数の研究で示唆されています。
