Application

ヒトジペプチジルペプチダーゼIVは、ヒトにおける相互作用的血行力学的阻害効果およびアンギオテンシン変換酵素阻害の研究に使用されています。ヒトジペプチジルペプチダーゼIVは同じく ハチ毒液アレルギーの解明について報告した研究でも使用されています。
シグマ製の酵素は DPP-IV阻害剤のスクリーニングおよび基質発見のためのLC-MS（液体クロマトグラフィ質量分析）ベースの分析方法を研究するために使用されています。

Biochem/physiol Actions

DPPIVには、プロリンの直後を切断するジペプチジルアミノペプチダ-ゼ活性があり、X-Pro-ZおよびX-Ala-Z配列を有するペプチドの非置換N末端からN末端ジペプチドを加水分解します。N末端残基Xは特に限定されず、またZはプロリン、ヒドロキシプロシン以外の残基を表します。
DPPIVは、X-Pro-ZおよびX-Ala-Zの配列を有する未置換のN-末端ペプチド由来のN-末端ジペプチドを加水分解するポストプロリンジペプチジルアミノペプチダーゼ活性を持っています。最適pHは7.4～8.7であることが分かっています。DPPIVは、免疫機能、炎症、 CNS 、内分泌機能、骨髄可動、癌の増殖、細胞接着、グルコース止血および敗血症／重篤感染などのいくつかの重要な生理学的プロセスの制御に関与します。[1][2][3]
天然のDPPIV は偏在性のII型膜貫通糖タンパク質で、S9 prolyloligopeptidaseファミリーセリンプロテアーゼです。in vivoでは、DPPIVは膜アンカードメインとして機能するシグナルペプチドを有した形で合成されています。 ヒト腎酵素とブタ腎酵素との間で、88%の配列相同性を示します。ヒト腎酵素とブタ腎酵素はともにサブユニット分子量約30 kDaのホモ二量体として存在します。 高マンノース 100 kDa DPPIV前駆物質は、ゴルジ体でプロセッシングされ、強固にN-結合およびO-結合した 124 kDaの成熟糖タンパク質となります。この成熟糖タンパク質は、N-結合グリカンおよびO-結合したグリカンと脂質ミクロドメインとの会合の協調作用を介して頂端膜に振り分けられます。ブタ酵素は炭水化物を18.3%含んでおり、グリカンの組成はフコース0.9%、マンノース3.4%、ガラクトース5.1%、グルコサミン8.2%、シアル酸0.7%です。 DPPIVは、内皮細胞、上皮細胞およびにリンパ球において高レベルで発現しています。また、血漿中に可溶性の形態で存在します。

    1. The volume in each vial can be calculated from information provided in the lot specific certificate of analysis found here:
      https://www.sigmaaldrich.com/product/sigma/d4943#product-documentation

      The specification is that each vial will be provided at a concentration of ≥10ug protein/mL, and that the activity will be ≥ 10units per mg protein. The calculations to make a solution of 0.005U/mL will be dependent on the specific activity of the lot.

As described above, DPPIV has a post-proline dipeptidyl aminopeptidase activity that hydrolyzes N-terminal dipeptides from the unsubstituted N-terminus of peptides with the sequence of X-Pro-Z and X-Ala-Z, where X is a nonspecific residue at the N-terminus and Z cannot be proline or hydroxyproline. pNA (p-nitroanilide) and AMC (7-amino-4-methylcoumarin) are substrate conjugates that are used for the quantitative analysis of enzyme activities by chromogenic or fluorogenic detection, respectively.
