ディスパーゼ I は、治癒プロセス初期における創傷サイズ上の羊水膜の効果を評価する研究において使用されています。 また、ディスパーゼ I は、プロテアーゼアッセイのためのジチロシン基質を調べる実験において使用されています。
Biochem/physiol Actions
ディスパーゼ I は、真皮からインタクトの表皮を分離できる、迅速で効率的、緩やかな中性プロテアーゼです。また、培養土台からインタクトの上皮シートを分離できます。本酵素は、上皮細胞の生存能力を保存したまま、基底膜部領域を切断します。また、懸濁培養における凝集の抑制にも使用できます。このプロテアーゼは、フィブロネクチンとタイプ IV コラーゲンを切断しますが、ラミニン、タイプ V コラーゲン、血清アルブミン、またはトランスフェリンは切断しません。[4]非極性アミノ酸残基の N 末端側のペプチド結合を加水分解します。また、露出した疎水性アミノ酸残基を持った、変性された細胞間のタンパク質を好んでアタックします。Ca2+、Mg2+、Mn2+、Fe2+、Fe3+ および Al3+ は、本酵素を活性化します。EDTA、EGTA、Hg2+ および他の重金属は、酵素活性を阻害します。[5]本酵素は、精製酵素 g-mol あたり 1g-原子の亜鉛を含んでいます。この亜鉛成分を、EDTA や EGTA などのキレート剤で除去した場合、不活性なアポ酵素が得られます。本酵素は血清によって阻害されません。
