General description

Amicon® Ultra-0.5 centrifugal filter devices provide fast ultrafiltration, with the capability for high concentration factors and easy concentrate recovery from dilute and complex sample matrices. For example, Cytochrome c (12.4 kDa) at 0.25 mg/ml initial concentration is typically retained at >95% .

The vertical design and available membrane surface area provide fast sample processing, high sample recovery (typically greater than 90% of dilute starting solution), and the capability for 30-fold concentration. Solute polarization and subsequent fouling of the membrane are minimized by the vertical design, and a physical deadstop in the filter device prevents spinning to dryness and potential sample loss. Efficient recovery of the concentrated sample (retained species) is achieved by a convenient reverse spin step after collecting the filtrate.

Amicon® Ultra-0.5 devices are supplied non-sterile and are for single use only. With their vertical membrane design and reverse spin, Amicon® Ultra 0.5 mL filters deliver great performance and lower spin times.

Application

  • Ultrafiltration with 10,000 Dalton molecular weight cutoff Ultracel® membrane
  • Concentration of biological samples containing antigens, antibodies, enzymes, nucleic acids, or microorganisms
  • Purification of macromolecular components found in tissue culture extracts or cell lysates, primer removal, linkers, or macromolecular labels from a reaction mix, and protein removal prior to HPLC
  • Desalting, buffer exchange, and protein dialysis

  1. What is the tolerance for molecular weight with these spin columns? Could you reasonably purify a protein of 15 kDa using these columns, or are the tolerances too high and would result in loss of protein into the filtrate?

    1. It is recommended to choose a molecular weight cut-off that is at least two times smaller than the molecular weight of the target molecules.

    1. This product is not suitable for separating protein/peptides of different sizes.

  3. If a centrifuge is unable to achieve a spinning speed of 14000 X g as recommended in the protocol, is it possible to compensate by spinning the samples for a longer duration?

    1. Yes, it is possible to compensate for a lower centrifuge spinning speed by increasing the duration. This can be achieved by calculating the K factor, which determines the adjustment needed in time.

  6. What is the maximum RPM that this can be spun to ensure the membrane does not perforate?

    1. The recommended G force for the Amicon® Ultra-0.5 filter is 14,000 g & 1,000 g reverse spin when using a Fixed-Angle (35 º) Rotor.
