General description

MCM7 is a highly conserved mini-chromosome maintenance protein (MCM) that is involved in the initiation of eukaryotic genome replication. It forms a hexameric complex with MCM4 and MCM6 (two subunits of MCM7, two subunits of MCM4 and two subunits of MCM7) and this complex regulates the helicase activity of the pre-replication complex. This complex is also involved in the formation of replication forks and in the recruitment of other DNA replication related proteins. MCM7 is reported to bind to DNA in metaphase II in oocytes and remains associated with the DNA until the S-phase. Two isoforms of MCM7 have been reported, 61 kDa and 90 kDa. In neuroblastoma tumors MCM7 expression is shown to correlates with MYCN (v-myc myelocytomatosis viral related oncogene, neuroblastoma derived) amplification where in MCM7 is rapidly up-regulated by Inducible MYCN Expression. MYCN protein is shown to bind specifically to the MCM7 promoter region, which possesses an E-box binding site for the Myc oncoproteins.
