General description

The SMC® Human Total-α-Synuclein High Sensitivity Immunoassay Kit contains all reagents required to perform the assay. This assay takes advantage of a standard immunoassay workflow configured in a standard plate format. Assay protocols are similar to existing sandwich ELISA methods with two key differences:
1) Elution buffer disrupts the sandwich, separating the labeled detection antibody for quantification.
2) The Erenna and SMCxPRO® immunoassay systems detect analytes using Single Molecule Counting (SMC®) technology.

Specificity

Species Reactivity Note

Specificity to the following species samples were tested with the following results:
Monkey – 100%
Cat – 100%
Rat – 100%
Mouse – 100%
Beagle – 100%

Cross Reactivity

Cross-reactivity to the following analytes were tested with the following results:
Tau – 0%UCHL1 – 0%
β-synuclein – 0%
γ-synuclein – 0%

Accuracy

CSF Recovery: 5 CSF samples spiked with average recovery of 96% and a range of 90-102%
Serum Recovery: 4 serum samples spiked with average recovery of 96% and a range of 93-98%
Plasma Recovery: 4 plasma samples spiked with average recovery of 96% and a range of 93-97%

Application

Uses 100 µL per well of 1:200 diluted serum or plasma samples, or 1:10 diluted cerebrospinal fluid (CSF) samples.
