Applicazioni

Quality Control Testing

Evaluated by Western Blotting in U-251 MG cell lysate.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected CYP46A1 in U-251 MG cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected CYP46A1 in lysate from human brain tissue and mouse brain tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CYP46A1 in human cerebral cortex tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CYP46A1 in U-251 MG cells.

Affinity Binding Assay: A representative lot of this antibody bound CYP46A1 peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.

Cholesterol 24-hydroxylase (UniProt: Q9Y6A2; EC:1.14.14.25; also known as CH24H, Cholesterol 24-monooxygenase, Cholesterol 24S-hydroxylase, Cytochrome P450 46A1) is encoded by the CYP46A1 (also known as CYP46) gene (Gene ID: 10858) in human. CYP46A1 is a member of the cytochrome P450 superfamily of enzymes. These enzymes are characterized by their heme-binding domain, which is crucial for their enzymatic activity involving electron transfer. CYP46A1 is a membrane-associated protein predominantly located in the endoplasmic reticulum. Its structure facilitates the oxidation of cholesterol, a critical step in cholesterol homeostasis and metabolism. The primary function of CYP46A1 is to catalyze the conversion of cholesterol to 24S-hydroxycholesterol. This conversion is a key mechanism for the elimination of cholesterol from the brain, as 24S-hydroxycholesterol can cross the blood-brain barrier, unlike cholesterol itself. This enzymatic activity is vital for maintaining cholesterol balance within the central nervous system and influences various cellular processes including membrane fluidity and the formation of lipid rafts. Alterations in CYP46A1 activity and expression have been implicated in several neurodegenerative diseases. Reduced activity of CYP46A1 has been associated with Alzheimer s disease, potentially due to impaired cholesterol turnover in the brain, which affects the generation and clearance of amyloid-beta plaques.

Stato fisico

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Stoccaggio e stabilità

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
