Specificità

Clone 1I7 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Thy-1 membrane glycoprotein (CD90). It targets an epitope within 19 amino acids from the internal region.

Immunogeno

KLH-conjugated linear peptide corresponding to 19 amino acids from the internal region of human Thy-1 (CD90) membrane glycoprotein.

Applicazioni

Quality Control Testing

Evaluated by Western Blotting in SK-N-SH cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Thy-1(CD90) in SK-N-SH cell lysate.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Thy-1(CD90) in K562 and TF-1 cell lysates.

Immunocytochemistry Analysis: A 1:1000 dilution from a representative lot detected Thy-1(CD90) in SK-N-SH cells.

Affinity Binding Assay: A representative lot of this antibody bound THY-1 (CD90) peptide with a KD of 2.1 x 10-7 in an affinity binding assay.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected Thy-1(CD90) in one million TF1 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Thy-1(CD90) in human tonsil tissue sections.

Anti-Thy-1(CD90), clone 1I7 ZooMAb®, Cat. No. ZRB1285, is a recombinant Rabbit monoclonal antibody that detects Thy-1 (CD90) and is tested in Affinity Binding Assay, Flow Cytometry, Immunocytochemistry, Immunohistochemistry, and Western Blotting.

Thy-1 membrane glycoprotein (UniProt: P04216; also known as CDw90, Thy-1 antigen, CD90) is encoded by the THY1 gene (Gene ID: 7070) in human. Thy-1 is a highly conserved, GPI-anchored membrane protein with one Ig V-type superfamily domain (aa 20-126). It is synthesized with a signal peptide (aa 1-19) and a propeptide (aa 131-161) that are subsequently cleaved off to generate the mature form (aa 20-130). It lacks an intracellular domain but is located in the outer leaflet of lipid rafts at the cell plasma membrane allowing signaling functions by cis- and trans-interactions with G inhibitory proteins, the Src and c-fyn kinases, and tubulin. Thy-1 is a major cell surface protein found on both T-cells and in the brain. It plays a key role in cell-cell and cell-ligand interactions during synaptogenesis and other events in the brain. It also plays a role in the regulation of axon growth and nerve regeneration, T cell activation and apoptosis, leukocytes and melanoma cell adhesion and migration, and fibroblast proliferation and migration in wound healing. Thy-1 mRNA and protein expression has been reported in several cancer types including liver, myeloid, skin, and brain. It is considered as a marker for glioblastoma stem cells. Reduced expression of Thy-1 is shown to enhance the expression of osteogenic and adipogenic differentiation of mesenchymal stem cells. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Stato fisico

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Ricostituzione

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stoccaggio e stabilità

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
