Signaling threshold-regulating transmembrane adapter-1 (SIT1) is a disulfide-linked homodimeric glycoprotein. It is mainly expressed in lymphocytes. The protein contains an amino-linked glycosylation site, transmembrane region and a cytoplasmic tail.

Applicazioni

The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. The antibodies that have been generated in support of the Tissue and Cancer Atlas projects have been tested by immunohistochemistry against hundreds of normal and disease tissues and through the recent efforts of the Human Cell Atlas project, many have been characterized by immunofluorescence to map the human proteome not only at the tissue level but now at the subcellular level. Azioni biochim/fisiol

SIT1 (Signaling threshold-regulating transmembrane adapter-1) inhibits T-cell receptor signaling and T-cell receptor-mediated phosphorylation of AKT (RAC-γ serine/threonine-protein kinase). Down-regulation of SIT1 promotes T-cell receptor-mediated AKT (RAC-α serine/threonine-protein kinase) activation and causes inactivation of FOXO1 (Forkhead box protein O1), a transcription factor that inhibits cell cycle progression and regulates T-cell homeostasis. SIT1 is involved in positive selection of T-cells. It regulates the signals required for thymic selection and peripheral T-cell activation.
