Application

Citrate Buffer, pH 6.0, 10×, Antigen Retriever has been used as a heat-induced antigen retriever on formalin-fixed paraffin-embedded (FFPE) tissue sections prior to application of antibodies or DNA fluorescent in situ hybridization.[1] In immunohistochemistry (IHC), most commonly used fixatives such as formalin mask tissue antigens (cellular, membrane, and nuclear) by their intrinsic crosslinking. This masking results in poor or no staining in IHC. The use of Citrate buffer, pH 6.0, or other antigen retrieval solutions on FFPE tissue sections improves accessibility of antibodies to tissue antigens.

Preparation Note

Dilute the Citrate buffer, pH 6.0, 10x, Antigen Retriever 10-fold with water to prepare a 1x Working Solution, e.g., dilute 10 mL of 10x concentrate with 90 mL of water.
Store the product at 2–8 °C. Do Not Freeze to prevent possible precipitation.

Other Notes

Occasionally the buffer may contain a yellowish tinge. This is due to preservative and will not affect the function of the buffer.
Store the prepared 1′ Citrate buffer, pH 6.0, Working Solution at 2–8 °C.

  1. Can you provide the composition of C9999-1000? Specifically, does it contain any detergents such as Tween?

  2. Is this a mixture from trisodium citrate and citric acid? I have looked through the SDS and the mixture of the chemical is <0.025% of 5-Chloro-2-methyl-4-isothiazolin-3-one and another chemical. I can't seem to find how they would make up a citrate buffer

    1. The specific formulation is considered proprietary, however, the final citrate concentration is 0.1M. The SDS lists only the potentially hazardous ingredients. The 5-chloro-2-methyl-4-isothiazolin-3-one and 2-methyl-2H-isothiazol-3-one (3:1) listed is essentially ProClin which acts as a preservative.   3. What molarity does this buffer have?

    1. Product C9999 is provided at a concentration of 0.1 M. It'll be 10 mM when diluted. The solution contains 116.25 mM Tri-NaCitrate, 10.25 mM EDTA, 0.5 % ProClin, and other proprietary materials.

  4. Is this Citrate buffer already included Tween 20?

    1. The composition of this buffer is considered proprietary. However, citrate solutions commonly used for antigen retrieval do not contain detergents. This product does contain a preservative which may contribute to the color of the solution. This does not effect the performance of the product in any way. Please see the link below to reivew a general antigen retrival protocol using Citrate Buffer. Note that Tween 20 is used in the wash solution following treatment with Citrate Buffer.
