應用

大观霉素是一种氨基环糖醇类抗生素,来源于 壮观链霉菌。它已被证明具有广泛的用途,包括治疗急性淋病性尿道炎和宫颈炎、标记细胞层以监测叶片发育期间的细胞命运、作为含有标记基因 Spcr 的植物相关转化系统中的选择标记、研究由 多杀性巴氏杆菌 溶血性曼氏杆菌 引起的牛呼吸道感染以及在 MS 琼脂培养基上产生质体编码 RNA 聚合酶缺乏的植物。大观霉素也可用于扩增携带低拷贝数的质粒。推荐在 7.5-20 mg/L 下用于细胞培养应用。

生化/生理作用

作用机制:壮观霉素通过与细菌30S核糖体亚基结合并干扰肽基tRNA易位而抑制蛋白质合成和延伸。

抗性机制:rpsE(核糖体蛋白S5的基因)突变可阻止壮观霉素的结合。氨基糖苷-3′-腺苷转移酶基因(aadA)也能使细菌产生抗性。

抗菌谱:壮观霉素能够对抗革兰氏阴性和革兰氏阳性菌。

注意

溶液可以在2-8°C下储存数周,或在-20°C下储存更长时间。

準備報告

制备储液时,应以10 mg/mL溶于水并过滤灭菌。
