Aplicación

El suero AB humano se utiliza en aplicaciones de ingeniería genética, trasplantes y terapia celular para la expansión de células madre mesenquimatosas (MSC) del tejido adiposo o las células del estroma mesenquimatoso de la médula ósea humana; para la generación y el trasplante de injertos normalizados de células madre del epitelio del limbo esclerocorneal; para la expansión ex vivo de células NK a partir de sangre periférica en medio de expansión de células madre hematopoyéticas; y para actualización de la tecnología de cultivo de islote humano pretrasplante.
El suero humano se ha utilizado como componente de la disolución salina tamponada con fosfato con detergente Tween® (PBST) en el enzimoinmunoanálisis de adsorción (ELISA) de doble captura, como muestra biológica simulada para evaluar el rendimiento del ELISA en papel con muestras biológicas complejas, como diluyente para crear una dilución en serie de la muestra basal para la generación de la curva de calibración para NISTmAb enriquecido en suero, así como para la muestra de paciente de mieloma múltiple (MM) MM2232

Acciones bioquímicas o fisiológicas

El suero AB humano se obtiene normalmente permitiendo la coagulación natural de sangre completa, lo que se conoce como suero «fuera del coágulo» (OTC). Se prefiere el suero humano obtenido de sangre completa coagulada, ya que en general contiene concentraciones mayores, y una gama más diversa, de factores de crecimiento para un crecimiento celular óptimo. El suero humano procedente de donantes de sangre de tipo AB carece de anticuerpos específicos para los antígenos de las sangres de tipo A y B. Esto lo convierte en un suero donante universal que es adecuado para varios tipos de cultivos celulares, entre ellas las líneas de células madre humanas.

Envase

El embalaje puede variar para diferentes lotes debido a la escasez de las botellas utilizadas para embalar este producto. La presencia o falta de envoltura retráctil alrededor de la tapa de la botella no afecta a la integridad/esterilidad del producto.

Advertencia

Se analizó cada donante para la hepatitis B y C y se observó que no eran reactivos , y tampoco lo eran para los anticuerpos del virus de la inmunodeficiencia humana (VIH) mediante ELISA. No obstante, los productos de origen humano deben considerarse potencialmente infecciosos y manejarse en consecuencia.

    1. The immunoglobulin concentration of this product is not measured. This is a pooled product from adult AB negative males. Based on information published in the Geigy Scientific Tables, the average immunoglobulin levels for adult males, ages 20 – 70, are as follows:

      1.32 – 2.34 g/L

    1. Each donor is tested for and found non-reactive to Hepatitis B & C and non-reactive to Human Immunodeficiency Virus (HIV) antibody by ELISA. HSV is not on the list of agents that are tested. Nevertheless, products of human origin should be considered potentially infectious and handled accordingly.   3. If this is "serum" derived from "plasma" the plasma would have contained anti-coagulant. To turn the plasma into serum it has to be clotted.  Has this product  been clotted to remove the fibrin? Or is it still in fact plasma?

    1. The product has been clotted. Blood is collected and treated with citrate (anticoagulant) which inhibits clotting initially. The blood is then centrifuged. When the spin is complete, layers will appear which allow a differentiation between the cells and the plasma. The plasma is removed and calcium is added to activate the clotting cascade. The preparation is then centrifuged again and the result is a large clot which is pelleted to the bottom of the tube with a serum layer on top.

    1. Human Serum products H4522 and H6914 may not be suitable for complement assays when stored at -20°C due to potential degradation of complement activity. For complement assays, alternatives like S1764 (Complement sera human) and C9473 (Complement Serum Standard human) are recommended. It is advised to review the Certificates of Analysis (CoAs) for these products to ensure they meet the specific requirements of your complement assay

    1. The use of human serum for a nucleic acid stability assay has not been performed as it is not part of our lab's product characterization focus.     1. Human AB serum is added as a supplement to culture media at a concentration of 2-10% to provide a mixture of nutritional, hormonal, growth, and attachment factors. The serum also acts as a buffer to the cell culture system against a variety of disruptions to cell growth and toxic effects such as pH change, proteolytic activity, or the presence of heavy metals or endotoxin.

  9. Is product H4522 obtained using Institutional Review Board (IRB)-approved consent forms and protocols?

    1. According to the supplier:
      Each donor unit used in the preparation of this material was tested and found negative for HBsAG, HBV-NAT, HIV-1/2, HCV, HCV-NAT, HIV-1 NAT, and Syphilis per FDA regulations. The human plasma was collected from male donors' blood type AB in FDA licensed commercial donor establishments located in the USA in accordance with ethical standards. Each donor signed an informed consent form prior to collection, which is traceable by the unique identifiable number to maintain anonymity. A quality assurance system is in place to ensure traceability to individual donor. Documentation to this effect is available upon request.

  10. Could you please confirm if the serum contains normal levels of human complement components?

    1. This product has not been treated to remove or inactivate complement components, however no testing has been performed to confirm these levels. It is a pooled product derived from 50 - 70 donors per lot. Any active complement be expected to meet 'normal levels.
