Especificidad

Clone 8H6.8 is a mouse monoclonal antibody that detects biotin-conjugated proteins.

Inmunógeno

Biotin conjugated to KLH.

Aplicación

Quality Control Testing

Evaluated by ELISA with Biotin-BSA.

ELISA Analysis: Serial dilutions from a representative lot detected Biotin-BSA in ELISA application.

Tested Applications

Western Blotting Analysis: A 1:5,000 dilution from a representative lot detected Biotin-BSA.

Immunofluorescence Analysis: A 1:50 dilution from a representative lot detected Biotin-conjugates in Human kidney tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Descripción de destino

Biotin, a bicyclic compound, is a water-soluble vitamin with a ureido (N-CO-N) and a tetrahydrothiophene ring. The ureido group is involved in its binding to avidin. The tetrahydrothiophene group is a valeric acid moiety. Although biotin molecule can exist in eight stereoisomers, the D-biotin isomer is the only one with biological activity. It plays important roles in a variety of critical metabolic reactions in the cell, and thus, is essential for normal human health, growth, and development. Its deficiency, particularly during pregnancy, leads to embryonic growth retardation, congenital malformation, and death. In mammals, biotin serves as a cofactor for four carboxylases that catalyze the transfer of a carboxyl group to targeted substrates. The four biotin-dependent carboxylases are: pyruvate carboxylase; propionly-CoA carboxylase; β-methylcrotonyl-CoA carboxylase; and acetyl-CoA carboxylase. These carboxylases play a critical role in the intermediate metabolism of gluconeogenesis, fatty acid synthesis, and amino acid catabolism. It is also reported to play a role in normal immune functions, including the production of antibodies, normal macrophage function, and the differentiation of T and B lymphocytes. This monoclonal antibody recognizes biotin conjugated to various proteins in ELISA and other immunochemical applications. (Ref.: Said, SM. (2012). Subcell. Biochem. 56; 1-19; Rabin, BS. (1983). J.
