General description

β-Estradiol or estradiol-17 β (E2) is secreted majorly by the large preovulatory follicles in the ovary. In addition, estradiol can be synthesized de novo from cholesterol have androgens synthesized from cholesterol. The aromatization of androgen by enzyme aromatase (P450arom) results in the β-estradiol production.

Application

β-Estradiol is used to study cell differentiation and transformations (tumorigenicity).
β-Estradiol has been used:
  • as a component of Dulbecco′s modified Eagle medium (DMEM) for culturing mammary tumor cells
  • as a oestrogenic compound for testing neuroprotective effects
  • as a component of medium199 for culturing oocytes

Biochem/physiol Actions

The major estrogen secreted by the premenopausal ovary. Estrogens direct the development of the female phenotype in embryogenesis and during puberty by regulating gene transcription and, thus, protein synthesis. It also induces the production of gonadotropins which, in turn, induce ovulation. Exposure to estradiol increases breast cancer incidence and proliferation.
The major estrogen secreted by the premenopausal ovary.
β-Estradiol (E2) is a major premenopausal hormone in women and is essential for spermiogenesis, sperm maturation and motility in men. Cultured bone marrow stem cells (BMSCs) have ability to synthesize E2 and could be exploited for treating estrogen deficiency. It mediates physiological functions through estrogen receptor (ER) α and ERβ.

Preparation Note

To prepare a 20 μg/ml stock solution, add 1 ml absolute ethanol to 1 mg β-estradiol; gently swirl to dissolve; add 49 ml sterile medium while mixing.

  1. Hello, you mention reconstitution in ug/ml. How much ethanol do I need to dissolve 250mg in to get a 100mM stock solution? Would I be able to do it directly in the glass bottle?

    1. To make a 100 mM stock solution with 250 mg of β-Estradiol (molecular weight 272.38 g/mol), dissolve it in 9.18 mL of ethanol. This is calculated as (0.250 g / 272.38 g/mol) / 0.1 mol/L = 0.00918 L or 9.18 mL. This can be done directly in a clean, dry glass bottle large enough to hold and mix the solution.

  2. Can Estradiol/ethanol solutions be sterile filtered?

    1. Yes. stock solutions in ethanol may be sterile filtered. However, cellulose-based filter membranes are not recommended for use with ethanol. Please see the link below to review additional information regarding filter membrane compatibility with commonly used solvents:

    1. The solubility of this product in DMSO has not been determined. However, as per the literature, the β-Estradiol is soluble in DMSO at 20 mg/ml.

  4. after dissolving the β-Estradiol in ethanol , whats the tempreture to store the item in ?

    1. The storage or stability conditions once the compound is in solution is not determined. General recommendations for β-Estradiol are to protect from light, and store at -20C or at 2-8C in a degassed solution. Solutions stored in this manner should remain stable in the range of 1 month - 1 year.

  5. How diluit for concentration of the 100ng/mL

    1. β-Estradiol is soluble in absolute ethanol at 50 mg/mL (See E8875). Preparing a 100 ng/mL solution would require serial dilutions.
      Start with a 1 mg/mL (1000 ug/mL) stock in ethanol.
      Dilute 10 uL (10 ug = 10,000 ng) of this stock with 9.990 mL to achieve a 1000 ng/mL solution. This can be diluted with medium or buffer.
      Dilute 1 mL of this with 9 mL to the final concentration of 100 ng/mL.
