Application

La dexaméthasone est utilisée pour étudier l'apoptose, les voies de signalisation cellulaire, la différenciation cellulaire[1] et l'expression génique.

Biochem/physiol Actions

Agent anti-inflammatoire du type glucocorticoïde
Agent anti-inflammatoire du type glucocorticoïde. Régule la survie, la croissance et la différenciation des lymphocytes T. Inhibe l′induction de l′oxyde nitrique synthase.
Ce produit est mal métabolisé par l'organisme et il présente une affinité et une biodisponibilité élevées.

Packaging

Flacon en verre à fond plat. Le produit se trouve dans un micro-insert conique à l'intérieur du flacon.

Physical form

Poudre à 2 °C à 8 °C ; à congeler en aliquotes de travail pour un stockage prolongé ; éviter les congélations/décongélations répétées.

Preparation Note

Pour préparer une solution mère à 20 μg/ml, ajouter un millilitre d'éthanol absolu par milligramme de produit ; mélanger jusqu'à dissolution en faisant délicatement tourner le flacon. Ajouter 49 ml de milieu stérile par millilitre d'éthanol ajouté, sous agitation, pour obtenir une concentration finale de 20 μg/ml.

    1. While we do not conduct in-house testing on these DMSO products in animals, there are numerous research papers available online documenting the utilization of this compound in animal studies. These papers serve as valuable resources for understanding the compound's effects and potential applications in various animal models:

    1. This product is stable for approximately 5 years in powder form with the vial unopened. It is guaranteed for 1 year from the date of shipment. The stability upon reconstitution has not been determined. However, for a similar product, it is recommended to aliquot and freeze at -20°C after reconstitution. Stock solutions are stable for up to 2 months at -20°C and should be protected from light. Degradation occurs gradually and depends on exposure frequency and intensity. Discolored, cloudy, or particulate-containing solutions should be discarded. Solutions can be sterile filtered using a 0.22 µm membrane, as the powders are not sterile.

  3. Hello,  I prepared a solution of Dexamethasone in DMSO and PBS for IP injection in mice for 20 days daily. Is it okay to store the solution at 4°C?

    1. Dexamethasone solutions in DMSO are stable for at least 30 days when stored at 4 °C and protected from light. Do not dilute to working concentrations in an aqueous buffer or medium until ready to use.

  4. Can I store D4902 at -80°C? If I store it at -80°C instead of -20°C, can I use it stably for more than 2 months?

    1. Long-term solution stability studies have not been performed on this material. The impact of stability at -80°C has not been determined. However, an analytical standard available at 1 mg/mL in methanol, as product D-085, has a shelf life of 4 years when stored at -20°C. In addition, therapeutic preparations of Dexamethasone 3.3 mg/mL for injection manufactured by Hospira, list a shelf life of 24 months at -20°C. This product, in solution, may demonstrate similar stability.

    1. The solubility of D4902 (Dexamethasone) in ethanol is 25 mg/ml and DMSO is 25 mg/ml. Dexamethasone is insoluble in water. Stock solutions are stable for up to 2 months at -20°C.

    1. Prepared solutions of Dexamethasone should be protected from light. Degradation occurs gradually and is dependent on the frequency and intensity of exposure. Solutions that appear discolored, cloudy, or contain particulates should be discarded. Unfortunately, there is not way to determine the degree of decomposition.

  7. I read that the dexamethasone is packaged in a bottomless glass bottle, in an inserted fused cone. Is this cone sealed with a rubber lid to be able to safely open the bottle and dissolve all contents before fully opening the bottle?

    1. Please see the attached images of the vial and cap used to package this product. The cap is a screw-type and does not include a septum. Threading secures the inner cone to the outer vial.

  8. what are different between D4902 and D1756? and if the same the both, why D4902 is descriptive about 'suitable for cell culture'?

    1. Product D4902 and D1756 are identical compounds, but are tested differently. Product D4902 is tested specifically for suitability in cell culture applications, while the D1756 has a slightly higher minimum purity specification.
