Especificidad

Clone 3F20 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects PALS1. It targets an epitope within 15 amino acids from the C-terminal half.

Aplicación

Quality Control Testing

Evaluated by Western Blotting in U2OS cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PALS1 in U2OS cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PALS1 in lysates from HeLa, and MDCK cells, and in Mouse brain and Rat brain tissue lysates.

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PALS1 in lysate from wild-type Caco2 and MDCK cells, but not in lysate from PALS1 knockout MDCK cells.(Data courtesy of Dr Michael Krahn from University Hospital of Munster in Germany).

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PALS1 in Human small intestine tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound PALS1 peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Protein PALS1 (UniProt: Q8N3R9; also known as MAGUK p55 subfamily member 5, Membrane protein palmitoylated 5, Protein associated with Lin-71) is encoded by the PALS1 (also known as MPP5) gene (Gene ID:  64398) in human. PALS1 is an evolutionary conserved Crumbs polarity complex protein that plays a KEY® role in the formation of apicobasal polarity and in tight junction biogenesis. It is also involved in adherens junction biogenesis by ensuring correct localization of the exocyst complex protein EXOC4/SEC8 that allows trafficking of adherens junction structural component CDH1 to the cell surface. It is shown to be essential during embryonic and postnatal retinal development and is required for the maintenance of cerebellar progenitor cells in an undifferentiated proliferative state, preventing their premature differentiation, potentially via promotion of mTOR signaling. Its expression is observed at the outer limiting membrane in the retina, kidney, and T lymphocytes. Mice that are functionally haploid for PALS1 develop a lethal phenotype, accompanied by heavy proteinuria and the formation of renal cysts. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity, reproducibility, and stability over conventional monoclonals. Forma física

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitución

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Almacenamiento y estabilidad

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
