General description

The kit is comprised of a combination of biological agents that reliably and completely eliminate mycoplasma contamination. The initial treatment of this eradication procedure is adequate for mycoplasma elimination in most applications. The second step suppresses and inactivates any remaining mycoplasma using a follow-up antibiotic treatment.

Application

The LookOut® Mycoplasma Elimination Kit has been developed to quickly and efficiently eliminate mycoplasma contamination from cell cultures.
The LookOut® Mycoplasma Elimination Kit is suitable for the elimination of Mollicutes and related organisms (Mycoplasma, Acholeplasma, Spiroplasma, and Entomoplasma) in cell and virus cultures. It is not effective for treating mycoplasma contamination in Chlamydia cultures.

  1. After I dilute my epithelial cells in the media + primary antibiotic, do I not change the media until they reach 80-90% confluency? I usually change the media on the cells every 2-3 days.

    1. The product information sheet protocol for this product instructs to grow the cells in the initial treatment media until 80–90% confluency before passaging. If a media change is necessary for cell health, it is important to include the elimination reagent in the new media.

    1. The LookOut Mycoplasma Elimination Kit is designed to efficiently eliminate mycoplasma contamination from cell cultures. This kit uses a combination of biological agents to reliably and completely remove mycoplasma contamination. The initial treatment is generally sufficient for mycoplasma elimination in most applications. A second step involves a follow-up antibiotic treatment to suppress and inactivate any remaining mycoplasma. The reagents have minimal cytotoxic properties, and the development of resistant strains is highly unlikely due to the significantly reduced mycoplasma concentration after the initial treatment. Compared to other products for mycoplasma elimination, inactivation, or suppression, the LookOut Mycoplasma Elimination Kit is more effective and has not been shown to cause changes in normal cell characteristics. It is the least cytotoxic method, with overall cytotoxicity demonstrated to be less than 20% across a diverse group of cell lines

    1. The key distinction lies in Product No. MP0030, LookOut® Mycoplasma Elimination Kit, which actively eradicates mycoplasma by disrupting the membrane. As a lipopeptide, it identifies cholesterol-rich regions in the membrane (as the concentration of cholesterol in mycoplasma membranes is higher than in eukaryotic cells), creating a pore through which the surrounding medium flows into the mycoplasma, ultimately causing it to burst. In contrast, standard antibiotics simply inhibit metabolic pathways, necessitating the rinsing out of mycoplasma through a medium change. If the process is halted prematurely, any remaining mycoplasma can regrow.
